LEGO bricks used as chemotatic chambers: evaluation by a computer-assisted image analysis technique

One of the main techniques used to explore neutrophil motility, employs micropore filters in chemotactic chambers. Many new models have been proposed, in order to perform multiple microassays in a rapid, inexpensive and reproducible way. In this work, LEGO® bricks have been used as hemotactic chambers in the evaluation of neutrophil random motility and chemotaxis and compared with conventional Boyden chambers in a ""time-response"" experiment. Neutrophil motility throughout the filters was evaluated by means of an image-processing workstation, in which a dedicated algorithm recognizes and counts the cells in several fields and focal planes throughout the whole filter; correlates counts and depth values; performs a statistical analysis of data; calculates the true value of neutrophil migration; determines the distribution of cells; and displays the migration pattern. By this method, we found that the distances travelled by the cells in conventional chambers and in LEGO® bricks were perfectly identical, both in random migration and under chemotactic conditions. Moreover, no interference with the physiological behaviour of neutrophils was detectable. In fact, the kinetics of migration was identical both in random migration (characterized by a gaussian pattern) and in chemotaxis (characterized by a typical stimulation peak, previously identified by our workstation). In conclusion, LEGO. bricks are extremely precise devices. They are simple to use and allow the use of small amounts of chemoattractant solution and cell suspension, supplying by itself a triplicate test. Reversal of visual impairment in patients with retinis pigmentosa after administration of glutathione peroxidase enzyme

The glutathione peroxidase enzyme (GPX) administration effects have been studied in patients with retinitis pigmentosa (RP). RP at present is defined as a degenerative, progressive and irreversible one. It was found that in RP patients there was a decrease of GPX level in respect to normal subjects. Moreover in rabbits it was shown that retrobulbar administration of GPX prevents the abolition of the electroretinogram due to previous administration of an oxidant agent as AgNO. On the basis of the results obtained, it was hypothesized that a deficit of reducing enzyme of the pentose phosphate pathway may binder the regeneration of the pigment modified by the oxidation process due to light that hits the retina. This process can bring to a pathological increase of the epithelial pigmentation. It was decided to investigate the potential effect of the replacement therapy with GPX administration. The number of patients is 7636 and their cumulative data are reported. The treatment was administration of one I.U. of GPX in retrobulbar fat, for three consecutive days each month, for three consecutive months, in each eye. We measured the visual acuity, the peripheral retinal fields and ERG before and after therapy. After completion of therapy over the 90% of the patients showed a significant clinical improvement, confirmed by the measured parameters. The amelioration for visual acuity ranged from 1/10 (R.E.) to no variation (L.E.) to 68% in both eyes. ERG, extinct in about 98% the patients, presented an improvement from a normal morphology with a very reduced voltage to a normal voltage and morphology. These results have been stable for the 6 years of follow-up. The results support the hypothesis that a deficit of GPX may play an important role in the pathophysiology of RP. These results conduct to consider that the pentose phosphate pathway is essential for the retina correct metabolism and function.

Tecniche di analisi non distruttiva su materiale lapideo

The thesis reports the activity performed in order to evacuate different kinds of sensors in material characterization and in anomalies detection. The paper is composed of an introductive section, containing a survey of NDT techniques used in various application, and of two sections containing experimental results. Section one describes the ideal response of a microwave resonant antenna used as a permittivity sensor, the data obtained by inspecting a set of homogeneous plastic samples with induced defects and a set of lapideous samples, and the data analysis. Section two reports the ideal response of a co-planar capacitor used as a permittivity sensor, the data obtained by inspecting the sets of samples and the data analysis. A comparison of the results obtained with the two sensor is reported: using the two techniques, complementary information regarding surface and proximal characteristics of the samples is obtained.

A study on vascular retinal alterations in patients with coronary artery disease

The paper describes the study carried out on a sample of 27 coronary artery disease patients of both sexes and with different agel aiming to determine a correlation between coronary artery disease and anomalies in the retinal circulation. Patients underwent selective coronary arteriography and fundus fluorescein angiography that allowed to investigate the dye dynamies and to detect abnormalities of time evolution, vessel walls and flux. In order to obtain a complete classifieation of the ischemie pathology of the retinal network we looked to the optic disc revolution, the papillo-macular network and the epi-peripapillar network. We have found a high evidence of abnormalities in all the examined sites; moreover we have observed a persistence of the dye in the ocular fundus longer than in people without coronary artery disease and we have found a correlation between the seriousness of the optie disc ischemia, c1assified in a numerical way, and the degree of the coronary artery disease. Hence the vascular abnormalities shown by the fluorescein angiography can constitute valid noninvasive markers and can advise further cardiologie tests for a coronary artery disease not yet diagnosed.

Studio delle paste di anfore della Laguna di Venezia : ausilio della elaborazione digitale di immagini

Presso l'ISDGM-CNR è in corso un progetto di ricerca archeometrica sulle ceramiche antiche che comporta lo studio archeologico e analitico di anfore iberiche importate nel Veneto durante il periodo romano. La scelta di questo materiale archeologico per gli studi sperimentali è stata suggerita dalla necessità di colmare una lacuna negli studi storici ma anche dalla supposta differenziazione archeometrica del materiale iberico e adriatico. Il progetto viene realizzato in fasi successive. Durante due anni di campagna di raccolta dei materiali scelti in circa quindici depositi in tutto il territorio veneto sono stati raccolti più di 300 campioni di anfore sia di tipo iberico sia di tipi diversi (adriatico-padano, greco-orientale e africano) destinati ad analisi comparative. Parallelamente è stato condotto uno studio archeologico che ha portato alla creazione di carte di distribuzione dei materiali studiati per tutta la zona, cioè terraferma, costa e laguna, che evidenziano alcune caratteristiche delle importazioni dei contenitori di alimentari nella antica Venetia; nel quadro di un programma comune dell'ISDGM-CNR e dell'Istituto di Archeologia e Etnologia (IAE-PAN) di Varsavia è stata inoltre verificata la stratigrafia dell'isola di Torcello. Conclusa la raccolta di materiali si è passati alla fase di studio e analisi dei risultati. La varietà dei problemi incontrati ha comportato l'impiego di vari metodi archeometrici e un approccio di studio interdisciplinare: sono state quindi attivate collaborazioni con diversi ambienti scientifici come l'Università di Pavia, il Centro Ceramico di Bologna, l'Università di Siviglia, l'AGH di Cracovia e, più recentemente, l'IEI-CNR di Pisa. 1 La collaborazione attivata con l'IEI ha come obiettivo lo studio di un metodo di analisi delle paste delle anfore che fornisca una classificazione oggettiva.

Elaborazione di fluorangiogrammi di pazienti con patologia arterio-coronarica per un'analisi correlata delle alterazioni retiniche

Un campione di 27 pazienti affetti da anomalie delle coronarie di diversa gravità è stato sottoposto alla osservazione oftalmologica. In generale, i soggetti esaminati hanno mostrato la presenza di anomalie del nervo ottico, del flusso epi-peripapillare e del flusso papillo-maculare: inoltre, nei soggetti del campione è stato misurato un ritardo nella comparsa del flush coroidale e una maggiore durata della fase lamellare, rispetto ai valori misurati in soggetti non coronaropatici. L'ischemia del nervo ottico è risultata di grado diverso a secondo dell'impegno coronarico segnalato; è stata inoltre riscontrata una correlazione tra le manifestazioni patologiche complessive riscontrate mediante l'indagine fluorangiografica e la patologia coronarica determinata mediante l'indagine coronarografica.

Studio di fattibilità per una stazione di acquisizione di immagini per fotogrammetria

Molte applicazioni in fotogrammetria richiedono elevate prestazioni di risoluzione spaziale, precisione e ripetitività di misura; il rilevamento numerico delle foto aeree dev'essere eseguito mediante l'impiego di una complessa struttura a controllo numerico, che comprende un'ottica di ripresa, un trasduttore opto elettronico ad alta risoluzione sia fotometrica che spaziale e vari componenti elettronici ed elettromeccanici: l'intera struttura dev'essere controllata mediante un'apposita procedura specializzata per l'ottimizzazione dell'efficienza operativa e di gestione. La digitalizzazione di un aerofotogramma comporta la produzione di una grande quantità di dati e richiede un tempo relati vamente lungo: può essere quindi conveniente realizzare una stazione dedicata prevalentemente alle operazioni di acquisizione dei dati, demandando ad altri sistemi di calcolo l'elaborazione delle immagini acquisite (v. fig. 1). La stazione di lavoro usata per l'elaborazione di immagini deve avere un'architettura tale da consentire il trattamento in tempi contenuti di grosse matrici di dati, mediante algoritmi anche complessi; la stazione di acquisizione può essere invece basata su un calcolatore monoutente di medie prestazioni, per es. un personal delle ultime generazioni, e deve avere un'architettura hardware e software, che consenta di eseguire in modo efficiente procedure predefinite per la gestione di dati. E' stato eseguito uno studio di fattibilita basato su questa ipotesi; in particolare sono stati presi in esame gli aspetti del problema piu rilevanti che possono condizionare l'efficienza operativa del sistema, occupandosi cioè di determinare alcune specifiche di progetto riguardanti l'acquisizione dei dati e la loro archiviazione.

A quantitative evaluation of the optic disc ischemia

The paper describes a method based on numeric techniques far image processing that can be used to determine the degree of the retinal ischemia. On the digital images obtained from fundus fluorescein angiography two regions of interest are selected and processed to obtain the ratio of photometric values of the two sets of pixels; this numerical ratio is used as an index of the disease. The semiautomatic method has been compared witb visual interpretation of fluorangiograms and a good agreement has been found. Relationship between coronary artery disease and retinal vascular alterations

Coronary artery disease (CAD) is frequently associated with peripheral vascular disease. In this study we investigated the relationship between CAD and retinal vascular abnormalities in 33 patients (26 men, 12 women, mean age 55±3 years) submitted to selective coronary angiography and retinal fluoroangiography. Coronary arteriograrns were evaluated to assess the presence and number of stenoses determining a lumen narrowing greater than 50% of the basal diameter. Digital quantitative analysis W:lS applied to retinal fluoroangiograms to determine the fluorescence-time curve, the presence of pcripapillar fluorescence and optic disc ischemia. The severity of optic disc ischemia also was evaluated semiquantitatively by assigning a score from 0 to 3. A 72-hour therapy washout was performed before the fluoroangiographic study, and patients with diabetes, systemic hypertension or heart failure were excluded from the protocol.

Studio delle paste ceramiche di anfore della laguna di Venezia mediante una procedura basata sulla elaborazione di immagini

The present article is part of a research project covering the imported and local amphorae found in the Veneto region. The study is specifically based on fragments of Spanish amphorae found in the Venetian lagoon. Several samples could be clearly identified as fragments of the forms coming from Baetica region (e.g., Dr 8, Dr 38, Dr 7/11, Dr 20 and Dr 20-23); other samples of very limited size could be reliably identified. In order to attempt a broad identification we have studied the composition of the pastes of a small number of fragments by means of mineralogical and petrographic analyses. In addition we used image processing techniques: in particular, we have designed a classification procedure that will perform morphologic, chromatic, ratiometric and spectral analyses on the images of thin sections of the amphorae, taken by a polarising microscope. The procedure could be completed by analysing a considerable amount of samples, that will allow to establish the decision thresholds used in the classification process; the procedure will facilitate the fusion of data and information obtained with different destructive and non destructive tests of the samples, so that it could constitute a useful tool for the archeological research.

An image processing procedure for the assessment of normality curves of motility of human granulocytes in micropore filters

With the conventional methods used in current practice, the results of human granulocyte motility assays are expressed by a numerical value that defines the leading front of cells into micropore filters. The authors have developed a fast procedure which allows the complete curve of polymorphonuclear leukocyte (PMN) migration to be obtained, from the initial plane of a filter to the maximum propagation depth. In this way, more information is given than that expressed by a single parameter. The paper describes the procedure used to determine the normality curves both for random and stimulated migration, according to a simple model which defines the PMN propagation in micropore filters.
